    Our Infini-D | Optimized Vitamin D Stack Story

    For many years we carried a vitamin D3 supplement that also included vitamin K2 to take advantage of the synergy between vitamin D and K. Over time it became clear there was also demand for a standalone vitamin D product. The problem was obvious: there are countless simple vitamin D supplements already out there, so we asked ourselves whether it was even worth releasing another basic D3 capsule. That question sent us down a long rabbit hole about how vitamin D actually works.

    The first order of business was to look deeper at the two common forms, D2 and D3. D3 (cholecalciferol) is derived from cholesterol and is the form we synthesize when UV light hits our skin. D2 (ergocalciferol) is derived from ergosterol, a similar compound found in fungi and plants that also produces the vitamin when exposed to UV light. The common rhetoric says D3 is simply superior, but the research is more nuanced than that. Both forms absorb equally well and both activate the vitamin D receptor with similar potency.

    What sets D2 and D3 apart, is that both forms have different pharmacokinetics. What's important to understand about vitamin D, is that both D2 and D3 are not active as is. They have to be converted to two different metabolites, the 25-hydroxy metabolites and then finally to the 1,25-dihydroxy metabolites. It's the 1,25-dihydroxy metabolites that mediate most of vitamin D's activity. This is where is gets interesting! D2 is harder to convert to its 25-hydroxy derivative and and once formed it is cleared more easily from circulation. Yet, it converts more efficiently to its active metabolite, 1,25-dihydroxy D2 (ercalcitriol), which is more metabolically stable and sticks around longer than 1,25-dihydroxy D3 (calcitriol). By combining both microencapsulated D3 and D2, we built a formulation that plays to the strengths of each rather than relying on a single form.

    To further round out the formulation, we added in a high efficiency butyrate precursor, Tributyrin. The reason it was added, is because butyrate has been shown to upregulate the enzyme that is responsible for generating the active 1,25-dihydroxy vitamin D metabolites. Furthermore, butyrate has also been shown to upregulate vitamin D receptor activity, which is very relevant here because most of vitamin D's benefits are mediated through the vitamin D receptor. We also added Maitake for its beta-glucan content, which adds another precursor for butyrate production. Maitake also contains ergosterol, the precursor for vitamin D2, which appears to be able to positively influence endogenous vitamin D3 production! Beyond this, Maitake is also a fantastic immune boosting mushroom which helps further enhance vitamin D's immune boosting properties. Last but not least, L-(+)-ergothioneine, a mushroom derived antioxidant compound which confers additional immune boosting properties whilst also helping to protect the vitamin D from oxidation.

    Mechanism of Action

    • Two Conversion Pathways: D3 and D2 are hydroxylated to their 25-hydroxy derivatives (calcifediol and ercalcidiol) and then to the active 1,25-dihydroxy metabolites (calcitriol and ercalcitriol), both of which activate the vitamin D receptor.
    • Metabolic Stability: Ercalcidiol converts efficiently to ercalcitriol, which is more metabolically stable and remains in circulation longer than calcitriol.
    • Ergosterol Contribution: Maitake mushroom extract supplies ergosterol, the same fungal precursor from which D2 is naturally derived.
    • Vitamin D Modulation: Tributyrin is a stable source of butyrate, which can upregulate CYP27B1, the enzyme that converts 25-hydroxy D3 and D2 into their active 1,25-dihydroxy forms, and can upregulate the vitamin D receptor (VDR).
    • Cellular Antioxidant Uptake: L-(+)-ergothioneine is taken up by a dedicated cellular transporter, concentrating it in tissues where it functions as a protective antioxidant.

    Vitamin D has been studied in the context of bone health. One trial found that vitamin D supplementation may help reduce wintertime and overall bone loss in healthy postmenopausal women [1], and another found that season and vitamin D status influence bone mineral density [2].
