What is Nicotinamide Mononucleotide (NMN Supplement | NAD+ Precursor)?

Nicotinamide Mononucleotide (NMN) is a derivative of Niacin. Niacin is a form of vitamin B3 and is also known as nicotinic acid. Vitamin B3 is one of 13 essential vitamins our bodies need. Nicotinamide Mononucleotide is known as a precursor to NAD+ (Nicotinamide adenine dinucleotide), NAD+ is a coenzyme in all living cells and plays a role in processes such as metabolism to DNA repair. Nicotinamide Mononucleotide is a critical component in the production of NAD+ because the body converts Nicotinamide Mononucleotide into NAD+. Since NAD+ levels are critical for keeping the mitochondria working at optimal levels, Nicotinamide Mononucleotide is viewed as one of the more important compounds in regards to maintaining cellular health.

There are a few differences between Nicotinamide Mononucleotide vs Nicotinamide Riboside. Nicotinamide Mononucleotide is not considered a form of vitamin B3 while Nicotinamide Riboside has shown to be a proven form of vitamin B3. In terms of structure, Nicotinamide Mononucleotide molecules are larger and weigh more than Nicotinamide riboside molecules. Nicotinamide Mononucleotide has shown positive effects on NAD+ levels in animal models and is currently being tested in humans.

Vitality with Nicotinamide Mononucleotide

Our cells’ ability to produce energy declines as we age. Although science isn’t sure why this happens, research has discovered that NAD+ levels decrease as we age which leads to the belief that lowered NAD+ levels are responsible for lower energy levels and that using a Nicotinamide Mononucleotide supplement may compensate for the loss of energy production.

Scientists ran several trials on the effects on mice after administering a Nicotinamide Mononucleotide supplement. Interestingly enough, energy promoting benefits were found exclusively in older mice. Research has found that Nicotinamide Mononucleotide benefits may help support overall cell proliferation. This leads to the belief that these cellular benefits are related to high NAD+ levels that maintain production of energy in the cell’s mitochondria.

Studies further found that regular NMN supplementation elevates NAD+ concentrations in the body, thus providing a readily available supply of NAD+ to be used. This research also demonstrated that NMN supplementation promotes the creation of NAD+ across organs such as the liver, muscle, kidneys, pancreas, blood vessels, and eyes. However, Nicotinamide Mononucleotide benefits go beyond increased NAD+ production. Some studies demonstrated that NMN may raise glucose tolerance and elevate insulin production under high fat diet conditions. This is believed to be the result of restoring sufficient NAD+ production and by balancing inflammation and stress caused by free radical damage. These studies found that NMN supports balanced inflammation levels within fatty tissue and improves sensitivity.

How Does NMN Absorb?

NMN has some very interesting absorption mechanisms, which leaves a lot of room for improving NMN bioavailability. In fact, there are even some rumors that NMN itself does not absorb at all, and has to be converted into Nicotinamide Riboside (NR) first. However, this rumor is false. NMN can absorb incredibly well by itself, but with a few caveats! For starters, NMN does not absorb in the stomach at all, and the compound is somewhat sensitive to the acidic environment in our stomach. NMN instead absorbs very rapidly and easily in the small intestines. This is due to the fact that NMN has its own specific transporter in the small intestines called Slc12a8.

Most compounds, including NMN, get absorbed through either passive diffusion mechanisms or active transporters. However, most of these active transporters are shared with other compounds, and can facilitate the transport of various different things. These different compounds then compete with each other for absorption through those transporters. For example, beta-alanine and taurine both share the same active transporter, and thus the two can compete for absorption. This is where NMN has a clear advantage! Since NMN has its own specific transporter, and if NMN makes it to the small intestines intact, it will be absorbed very efficiently. This is because there are no other compounds that can compete with NMN absorption through the Slc12a8 transporter. The trick here is getting NMN into the small intestines intact, and this is where an enteric coating comes in and helps to enhance overall NMN bioavailability!

Enteric Coating For Better NMN Bioavailability

Since NMN only absorbs in the small intestines through the Slc12a8 transporter, the best NMN bioavailability can be achieved by ensuring safe passage through the stomach and into the small intestines. NMN may degrade in the stomach due to the acidic conditions, thus we decided to put an enteric coating on our NMN tablets. This enteric coating prevents disintegration of the NMN tablet in the stomach and instead delays tablet disintegration until the NMN tablet reaches the small intestines. By utilizing the enteric coating, we can ensure the highest possible concentration of NMN in the small intestines, thus optimizing NMN bioavailability.

    Appears to be what it says on the label. I’m not really the target audience but I was curious if it’s effects. Shortly after taking it I feel a bit warm all over, but that’s about it. It’s clearly doing... something, but whether or not it’s helpful... I guess only time can tell.

    First off, I’ll just say that many nootropics do not work for me unfortunately. I am a 28yo mutant that wishes to improve my health in any way I can. I have low energy and was hoping that this NMN would give me a good push. Long story short, it makes me sleepy or tired for some reason. Maybe I’m too young? This is from 125mg, so idk if taking 2 125mg would make a difference. ND Amdin, do you have any idea why this could be possible? Do you have any recommendations?? ND Admin: NMN shines when combined with other compounds that affect mitochondrial energy and AMPK. So combing it with CoQSol-CF, berberine, EGCG, hesperidin, OptimALA, etc. is a good idea. However, not everything will be good for everyone. You might also have high CD38 levels, which is preventing the NMN from becoming NAD+. So taking something that inhibits CD38 would be good. I would suggest quercetin for that. So adding in CoQSol-CF, hesperidin, quercetin, and EGCG would be a good place to start.

    I love three things about this product. First, you can cut a lot of products of your skin, sleep, and health stacks because NMN already helps a lot in these areas. The second one is that you can recommend this product to many people without having to explain how to take it, talk about side effects, etc. Finally, it's probably the best substance available to buy, and ND offers, by far, the absolute best one.

    Been taking it twice a day and about a week in, I noticed a difference. I have partied hard and fast in the past and just turned 30 this year. I've been trying to take better care of myself and really I'm on a good regimen with some good supplements and amino acids, but this is one that I wish I would have added sooner. It's crazy how much more I feel like "myself" again. I have energy, I basically feel like I did when I was 18 years old. Outgoing again, minimal anxiety, sleeping excellent, almost feeling high on life. It is truy incredible. And I attribute it to this. Be sure to take methyl donors like methylfolate, TMG, methylcobalamin (methyl-B12) , etc cuz any niacin derivatives can reduce ur methyl donors. I take high dose Methylfolate 10-15mg a day since I have MTHFR , and an active B Complex , along with some others and have no problems! All about balance! ND recommends combos like NMN & Citicoline, NMN & COQ-10, ND with Ashwagandha, and I use em all! Been about a month since I started. Thank God N.D. came out with their version of this product that's of high quality and delicate handling from very beginning. Would not have thought it was such an unstable substance, especially when other companies sell it in capsules. Jumped at the chance when it came out. Lifetime user! Another win for N.D. they're truly the top of the game :)

    Wasn't sure what to expect. I'm in my mid twenties and this is generally recommended for those over thirty. I was incredibly surprised by my results! My skin has never looked better, my energy remains consistent throughout the day, and my willingness to engage in activities I normally find undesirable has improved. These effects are compounded when taking it twice a day, but once a day still provides plenty of benefits. I wouldn't trust another companies NMN. Their price is competitive and their quality unparalleled. I highly recommend this as a base supplement for any nootropics stack.

    The first few days, the effects were quite mild, but after taking it for a week, the effects have now become very noticeable. I get a great lift in energy levels, especially when I take it alongside CoQsol-CF. I also think it has started to improve my skin quality and I feel like my sleep has become more restful. Overall, I think the hype that NMN gets on various forums I researched it on, is well deserved and I'm excited to see if the effects will keep getting better with time!
