Sarcosine is also known as N-Methylglycine and Methylglycine. Sarcosine is synthesized from the glycine N-Methyltransferase (GMNT) enzyme which utilizes a methyl group from S-Adenosyl methionine and donates it to glycine. This process creates Sarcosine and S-adenosylhomocysteine.
Sarcosine shares properties with glycine and D-serine but its effects are weaker. However, sarcosine is more reliably absorbed by the body than D-serine so a smaller more practical dose of sarcosine would achieve the same effects compared to glycine or D-serine.* Sarcosine metabolism is achieved through pipecolic acid oxidase (PIPOX) or sarcosine dehydrogenase (SARDH) enzymes. PIPOX and SARDH convert sarcosine into glycine.
Sarcosine has binding capability to NMDA (N-Methyl-D-aspartate) receptors. Research has found that Sarcosine seems to promote feelings of well being and support cognition by enhancing NMDA receptor signaling via inhibiting one of the major glycine transporters.
Glycine transport inhibitors such as Sarcosine block the uptake of glycine, leaving more glycine present in the synapse. As a result, NMDA receptor signaling is notably enhanced. GlyT1 and GlyT2 are two of the main glycine transporters. Sarcosine targets the GlyT1 transporter which is the more prominent transporter expressed on glial cells. GlyT1 has been observed to be more involved with NMDA signaling than GlyT2. GlyT1 inhibitors such as Sarcosine have also been observed to have a positive effect on cognition but further research is needed to gain a firm conclusion.
While there is still research that needs to be done to determine all the beneficial effects of taking a sarcosine supplement, there are a few isolated human studies that had interesting findings. In one human study on Sarcosine, it was found that subjects self reported increased motivation and less irritable mood when supplementing with 1g-2g of sarcosine powder.
As a dietary supplement, take one 1000mg Sarcosine tablet 1-2 times daily. For milder effects, break tablet in half.
