Nicotinamide Mononucleotide (NMN) is a derivative of Niacin. Niacin is a form of vitamin B3 and is also known as nicotinic acid. Vitamin B3 is one of 13 essential vitamins our bodies need. Nicotinamide Mononucleotide is known as a precursor to NAD+ (Nicotinamide adenine dinucleotide), NAD+ is a coenzyme in all living cells and plays a role in processes such as metabolism to DNA repair. Nicotinamide Mononucleotide is a critical component in the production of NAD+ because the body converts Nicotinamide Mononucleotide into NAD+. Since NAD+ levels are critical for keeping the mitochondria working at optimal levels, Nicotinamide Mononucleotide is viewed as one of the more important compounds in regards to supporting cellular health.
There are a few differences between Nicotinamide Mononucleotide vs Nicotinamide Riboside. Nicotinamide Mononucleotide is not considered a form of vitamin B3 while Nicotinamide Riboside has shown to be a proven form of vitamin B3. In terms of structure, Nicotinamide Mononucleotide molecules are larger and weigh more than Nicotinamide riboside molecules. Nicotinamide Mononucleotide has shown positive effects on NAD+ levels in animal models and is currently being tested in humans.
Our cells’ ability to produce energy declines as we age. Although science isn’t sure why this happens, research has discovered that NAD+ levels decrease as we age which leads to the belief that lowered NAD+ levels are responsible for lower energy levels and that using a Nicotinamide Mononucleotide supplement may compensate for the loss of energy production.
Scientists ran several trials on the effects on mice after administering a Nicotinamide Mononucleotide supplement. Interestingly enough, energy promoting benefits were found exclusively in older mice. Research has found that Nicotinamide Mononucleotide benefits may help support overall cell proliferation. This leads to the belief that these cellular benefits are related to high NAD+ levels that maintain production of energy in the cell’s mitochondria.
Studies further found that regular NMN supplementation promotes NAD+ concentrations in the body, thus providing a readily available supply of NAD+ to be used. This research also demonstrated that NMN supplementation supports the creation of NAD+ across organs such as the liver, muscle, kidneys, pancreas, blood vessels, and eyes. However, Nicotinamide Mononucleotide benefits go beyond NAD+ production. Some studies demonstrated that NMN may promote glucose tolerance and insulin production under high fat diet conditions. This is believed to be the result of restoring sufficient NAD+ production and by balancing inflammation and stress caused by free radical damage. These studies found that NMN supports balanced inflammation levels within fatty tissue and improves sensitivity.
NMN has some very interesting absorption mechanisms, which leaves a lot of room for improving NMN bioavailability. In fact, there are even some rumors that NMN itself does not absorb at all, and has to be converted into Nicotinamide Riboside (NR) first. However, this rumor is false. NMN can absorb incredibly well by itself, but with a few caveats! For starters, NMN does not absorb in the stomach at all, and the compound is somewhat sensitive to the acidic environment in our stomach. NMN instead absorbs very rapidly and easily in the small intestines. This is due to the fact that NMN has its own specific transporter in the small intestines called Slc12a8.
Most compounds, including NMN, get absorbed through either passive diffusion mechanisms or active transporters. However, most of these active transporters are shared with other compounds, and can facilitate the transport of various different things. These different compounds then compete with each other for absorption through those transporters. For example, Beta-alanine and Taurine both share the same active transporter, and thus the two can compete for absorption. This is where NMN has a clear advantage! Since NMN has its own specific transporter, and if NMN makes it to the small intestines intact, it will be absorbed very efficiently. This is because there are no other compounds that can compete with NMN absorption through the Slc12a8 transporter.
