P5P plays a major role in neurotransmitter synthesis but we need to cover some steps to show how it does this! Vitamin B6 plays a major role in the metabolism of amino acids through decarboxylation, transamination, deammation, racemization and desulfhydration reactions. This is very important because most neurotransmitters are derived from amino acids. For example, serotonin is synthesized from the amino acid L-Tryptophan. L-Tryptophan, via the enzyme tryptophan hydroxylase, is converted to 5-hydroxytryptophan (5-HTP). 5-HTP is then converted in a single step to serotonin by another enzyme called aromatic L-amino acid decarboxylase (AADC). AADC depends on P5P, which means that without P5P, you cannot complete the last step in serotonin synthesis. This very same enzyme, also generates the following neurotransmitters and neuromodulators
- Dopamine
- Histamine
- Phenethylamine
- Tyramine
- Tryptamine
This means that without P5P, any neurotransmitter or neuromodulator that is synthesized by AADC, will not be produced.
In addition to AADC, another enzyme called glutamic acid decarboxylase (GAD), is heavily dependent on P5P. GAD converts the excitatory amino acid neurotransmitter glutamate to the inhibitory amino acid neurotransmitter GABA. This means that P5P is also crucial for maintaining the balance between glutamate and GABA in the brain.
Finally, P5P also regulates prolactin levels, a hormone which also has a close relationship with dopamine. When dopamine levels drop, prolactin levels can increase which can have undesirable physical and mental effects. This is where P5P can really shine, as it both limits the release of prolactin, while also enhancing dopamine levels!
